Bianca Brundel is moleculair en cellulair bioloog en hoogleraar moleculaire biologie van boezemfibrilleren. Brundel is bekend om haar experimentele (dierproefvrije) modelsystemen die aangrijpingspunten voor de behandeling van boezemfibrilleren laten zien.[1] Brundel is werkzaam in het Amsterdam UMC en is co-founder van het boezemfibrilleren co-creatie platform Stichting AFIP.
Jeugd en Opleiding
Bianca Brundel groeide op in Lichtenvoorde en bezocht het Marianum te Groenlo. Daarna is ze de opleiding biochemisch en biotechnologisch laborant aan de hoge laboratorium opleiding in Enschede gaan volgen, gevolgd door een master opleiding moleculaire en cellulaire biologie aan de Vrije Universiteit Amsterdam. Tijdens haar masteropleiding heeft Brundel zich extra gespecialiseerd in de industriële genetica en populariseren van de biologie.
Carrière
In 1995 is Brundel haar promotieonderzoek gestart bij de afdeling Klinische Farmacologie en Cardiologie UMC Groningen onder begeleiding van Isabelle van Gelder, Harry Crijns, en Rob Henning. Voor dit onderzoek heeft Brundel de moleculaire en structurele veranderingen in menselijk boezem hartweefsel in kaart gebracht van patiënten met boezemfibrilleren ten opzichte van patiënten zonder boezemfibrilleren. Haar belangrijkste bevindingen zijn de afbraak van specifieke eiwitten (myolyse) door boezemfibrilleren. Dit heeft voornamelijk betrekking op zogenaamde ionkanalen[2] en eiwitten[3] die betrokken zijn bij het samentrekken van de boezemhartcel. Een speciek eiwit, calpaine, is betrokken bij de eiwitafbraak en structuur veranderingen in de boezem. Een behandeling gericht op het voorkomen van myolyse[4] zou boezemfibrilleren helpen voorkomen in patiënten. Om dit concept verder uit te werken is Brundel gaan werken bij de afdeling Celbiologie, UMCG (Prof. Harrie Kampinga) en aan de Montreal Heart Institute (Prof. Stanley Nattel[5], Canada).
Na haar terugkeer in Nederland is Brundel zich verder gaan specialiseren bij de afdeling Klinische Farmacologie en sinds 2016 is ze werkzaam als hoogleraar bij de afdeling Fysiologie, Amsterdam UMC, VUmc, Amsterdam Cardiovascular Sciences.
Denkrichting en erkenning
De studies van Brundel zijn vooral gericht op het ontrafelen van de moleculaire mechanismen, die ten grondslag liggen aan boezemfibrilleren. Centraal in haar onderzoek staat de rol van het ontsporen van de eiwit homeostasis, ookwel proteostasis[6] genoemd. Belangrijke mechanismen voor proteostasis ontsporing in boezemfibrilleren zijn: het uitputten van de beschermende heat shock proteinen (HSPs)[7], overactivatie van autofagie[8], afbraak van het microtubuline netwerk[9] en het ontstaan van DNA schade[10].
Brundel werkt hoofdzakelijk met zelf opgezette experimentele modelsystemen voor boezemfibrilleren, namelijk: gekweekte boezem hartcellen [4] en Drosophila[11]. Door experimentele uitkomsten te vergelijken met de bevindingen in menselijk hartweefsel, geeft Bianca haar onderzoek een translationeel karakter. Samen met Natasja de Groot[12] (Erasmus MC), test zij nieuwe geneesmiddelen en voedingssupplementen gericht op het herstellen van de schade in boezem hartcellen in en in samenwerking met patiënten met boezemfibrilleren. Voorbeelden studies zijn de Glutaminimize AF[13] studie en GGA studie[14].
De inzichten van Brundel zijn in 2022 opgenomen in Nature Reviews Disease Primers ‘Atrial Fibrillation’[15] en hebben geresulteerd in het verkrijgen van een subsidie binnen de Nationale WetenschapsAgenda (2022), project CIRCULAR[16], waarin Stichting AFIP een centrale rol speelt.
Project CIRCULAR
Brundel heeft de leiding over het project CIRCULAR. Binnen het onderzoek naar triggers en voorkomers van boezemfibrilleren, staan de ervaringen van patiënten centraal. Deze ervaringen zijn verzameld via stichting AFIP en samen meten diverse onderzoekers (waaronder Natasja de Groot, Peter van Tintelen, Umut Konus, Yannick Taverne, Esther Creemers en Martin Bennink) richt dit project zich op het in kaart brengen van de moleculaire mechanismen en hoe triggers boezemfibrilleren veroorzaken of stoppen.
Triggers die Brundel vanaf het najaar van 2022 onderzoekt zijn onder andere: stress, genetische variatie en pesticiden.[17][18] Daarnaast onderzoekt zij potentiële remmers van boezemfibrilleren, waaronder: muziek en een ‘whole foods plant-based diet’.
